The Oxford adenovirus COVID-19 vaccine is safe and elicits significant immune responses in study volunteers, according to research published in The Lancet.

Among the myriad of vaccine candidates that researchers across the globe are developing to stop the spread of the new coronavirus, SARS-CoV-2, the Oxford vaccine has already garnered significant attention.

The vaccine, which the researchers call ChAdOx1 nCoV-19 but many refer to as the Oxford vaccine, uses a weakened adenovirus from chimpanzees.

Together with colleagues across the United Kingdom and Europe, the researchers from the University of Oxford in the U.K. previously demonstrated that the vaccine could protect rhesus macaques on subsequent exposure to the new coronavirus. This research has not been through full peer review yet.

A further study, which has also not undergone peer review yet, indicated that pigs were able to produce neutralizing antibodies to SARS-CoV-2, in addition to showing T cell responses. A second booster shot led to an enhanced antibody response, the authors write.

The latest results in The Lancet show that the vaccine is safe in humans and that it also produces a measurable immune response. Experts have reacted with cautious enthusiasm to the preliminary findings.

For their study, the researchers recruited 1,077 volunteers at five study sites across the U.K., half of whom received the Oxford vaccine while the other half received a meningitis vaccine, which served as the control.

The average age of the study participants was 35, the gender split was roughly equal, and 90.9% were white.

Side effects in response to the Oxford vaccine were common but generally mild. They included fatigue, headaches, pain at the injection site, and flu-like symptoms. Prophylactic acetaminophen, which a subset of volunteers received, was effective in reducing these symptoms.

The quantity of antibodies that the team measured peaked 28 days after injection and remained elevated at 56 days.

The 10 participants who received a booster shot of the Oxford vaccine had an increased number of antibodies at 56 days.

When the researchers specifically looked for neutralizing antibodies, which can stop the virus from infecting host cells, they found that the majority of people had developed these.

The team used several different laboratory assays to screen for these antibodies, showing that all of the people who had received the booster had neutralizing activity. Depending on the assay, 62–100% of people who had received just one dose had neutralizing antibodies.

However, antibodies are not the only way in which people can fight off pathogens. T cells, which are a type of white blood cell, can attack cells that are infected with a virus.

The team was able to measure T cell responses within 7 days of vaccination. These peaked at 14 days and were still measurable at 56 days. Delivering a second booster shot had no effect on T cell responses.

While these results certainly are encouraging, the study authors highlight some limitations. These include the short follow-up time and the fact that most of the study participants were young, white, and in good health.

“Further studies are required to assess the vaccine in various population groups, including older age groups, those with comorbidities, and in ethnically and geographically diverse populations.”

Phase III trials are currently underway.

Reactions to the results have been positive, but experts do urge caution.

“We are still in the early days of understanding what constitutes an effective long-term immune response against SARS-CoV-2,” says Dr. Doug Brown, Chief Executive of the British Society for Immunology. “This paper shows that the Oxford vaccine elicited both neutralizing antibodies and T cell immune response against SARS-CoV-2 in the majority of patients after a single dose — this is a good first sign, but we still have yet to understand whether these immune responses will have an effect if those individuals encounter the SARS-CoV-2 virus in the future.”

“It has encouraging results, but it is not the end of the road, far from it, but a vital, necessary step toward an effective vaccine without significant harms,” comments Prof. Stephen Evans, a professor of pharmacoepidemiology at the London School of Hygiene and Tropical Medicine in the U.K.

“The key elements required to proceed to a phase III trial are all there. The responses measured in the blood and the absence of serious harms indicate there is a possibility of an effective vaccine against COVID-19,” he continues. “It does not yet show that the disease is reduced or prevented, and this will not be easy to show until phase III trials have been completed in settings where the SARS-CoV-2 virus is circulating at a high rate, and people are getting clinical and severe disease.”

The authors also take a cautionary approach to their conclusions. “Further clinical studies, including in older adults, should be done with this vaccine,” they write about the implications of the evidence of their work.

Promising results from trials of three potential coronavirus vaccines have raised hopes in the fight against the COVID-19 pandemic, which has killed more than 600,000 people and crushed economies globally.

Early-stage clinical trials of AZD1222, a closely-watched experimental vaccine being developed by AstraZeneca and scientists at Oxford University in the United Kingdom, induced “robust immune responses” in participants without any serious side-effects, according to results published in The Lancet medical journal on Monday.

The Oxford vaccine is one of 150 in development globally, but is considered the most advanced. US drugmaker Pfizer and China’s CanSino Biologics also reported positive responses for their candidates on Monday

The World Health Organization (WHO) has said any viable vaccine must be made available to everyone.

Below, we take a look at the latest developments in the global vaccine race.

What is AZD1222?

The AZD1222 vaccine has been adapted from a common cold virus found in chimpanzees, with spike glycoprotein, a genetic material from the new coronavirus, added to it.

It uses the modified virus to deliver genetic instructions to cells to induce an immune response against the novel coronavirus that causes COVID-19.

Between April and May, a total of 1,077 healthy male and female adults between the age of 18 and 55 in five UK centres took part in the randomised and controlled Phase I and II trials of the experimental vaccine.

The participants were each given one dose and followed for about a month. Ten of them were given a second dose after 28 days.

What did the results show?

In its Phase I trial, the vaccine induced so-called neutralising antibodies – the kind that stop the virus from infecting cells – in 91 percent of individuals one month after they got one dose, and in all 10 subjects who got a second dose after 28 days, echoing a similar trial in pigs.

These levels were on par with the antibodies produced by people who had survived a COVID-19 infection – a key benchmark of potential success.

Oxford lead researcher Sarah Gilbert said the trial could not determine whether one or two doses would be needed to provide immunity.

The vaccine also induced the body to make T cells – activating a second part of the immune system that experts increasingly believe will be important for a lasting immune response.

What is the reaction?

While welcoming the early results, the scientific community also urged caution, with some predicting it would take until the middle of next year to find out if the vaccine actually works.

Andrew Pollard, chief investigator of the Oxford study, said they were at “an important milestone” in the anti-coronavirus efforts but added the researchers were “now moving rapidly forwards to try to evaluate whether the vaccine actually protects the population by conducting large-scale trials”.

Mene Pangalos, AstraZeneca’s research chief, said in a statement that Monday’s data “increases our confidence that the vaccine will work and allows us to continue our plans to manufacture the vaccine at scale for broad and equitable access around the world”.

Mike Ryan, head of emergencies at the WHO, praised the “good news” from the results, but warned: “There’s a long way to go.”

UK Prime Minister Boris Johnson, whose government has helped to fund the project, hailed the announcement as “very positive news”.

What next?

Before a vaccine can be approved for mass production and global distribution, it has to go through three stages, sometimes four, of clinical trials in humans. Provoking an immune reaction is the first part of vaccine development.

The Phase III trials, already under way in the UK, South Africa and Brazil, are looking at the optimal dosage and how much protection vaccinated people have when actually exposed to the coronavirus. The late-stage trials are also due to start in the United States, where prevalence is higher.

“There’s still a long way to go,” Gilbert, the Oxford researcher, said.

“We still need to see how the vaccine performs in older people, who are more risk of severe disease than the people we’ve vaccinated in this study,” she said. “So that’s the subject of future work, and there’ll be more publications to come.”

Wafaa el-Sadr, professor of epidemiology and medicine at Columbia University, told Al Jazeera a timeline of six months would likely determine if the vaccine is safe and effective for mass production.

AstraZeneca has signed agreements with governments around the world to supply the vaccine, should it gain regulatory approval. It has signed deals to produce and supply more than two billion doses of the shot, with 300 million doses earmarked for the US.

What other vaccines are being developed?

More than 150 other vaccines against COVID-19 are being developed around the world.

New data released on Monday showed that a candidate developed by CanSino Biologics Inc and China’s military research unit appeared to be safe and induced an immune response in most of the 508 healthy volunteers aged 18 to 83 who received one dose of the vaccine.

Some 77 percent of study volunteers experienced fever, fatigue, headache or pain at the injection site, side-effects not considered to be serious.

Results of another, smaller study in Germany of a different type of vaccine that uses ribonucleic acid (RNA) – a chemical messenger that contains instructions for making proteins – were also released by German biotech company BioNTech and Pfizer.

In the study of 60 healthy adults, which was not peer-reviewed, the vaccine induced virus-neutralising antibodies in those given two doses, a result that was in line with a previous early-stage US trial.

While the pace of the global vaccine race has been unprecedented, experts caution that producing enough doses of a successful candidate may be as difficult as developing the candidate in the first place.

A senior British official has called the latest news on an Oxford University team’s potential coronavirus vaccine “extremely encouraging.” The results of the Phase I/II trial of the vaccine being developed by Oxford’s Jenner Institute, in conjunction with pharmaceutical giant AstraZeneca, showed that it is safe and “produced strong immune results,” according to the research published on Monday in The Lancet medical journal.

The vaccine caused a 2-pronged immune response, a news release from the Jenner Institute at Oxford said. First, within 14 days, it triggered a T cell response, generating white blood cells that can attack infected cells. Second, within 28 days, it provoked an antibody response. Antibodies are able to prevent the virus from infecting cells when it is initially contracted, according to the release.

“Those are two parts of the immune system that ideally work together to protect against viral infections,” Professor Sarah Gilbert, who heads up the Oxford team, explained to CBS News’ Charlie D’Agata.

Crucially the trial also showed that the candidate vaccine produced no serious side effects, causing only minor headaches, fatigue and some soreness around the point of injection. 

“We’re all getting a sigh of relief,” CBS News medical contributor Dr. David Agus told D’Agata of the trial results. “We’re getting the immune responses that science hoped for.

Oxford is working with AstraZeneca to develop, manufacture, and produce a coronavirus vaccine on a large scale. The unprecedented effort aims to make some 2 billion doses of the vaccine available globally, through partnerships with manufacturers in several countries, by early next year. 

Lead scientist Gilbert told the BBC on Tuesday morning that while the hope was still to be able to make millions of doses available by this autumn, there was still “absolutely no certainty” of that timeline, as the final, Phase III trials were still ongoing, and the roll-out of the vaccine on a large scale was also contingent on the manufacturing process and individual nations’ safety regulators approving it for emergency use.   

While the latest results are encouraging, they show that the trial vaccine creates the reaction in the body that should give immunity to the disease. The Phase III trials, to prove it actually gives people that immunity in the real world, are already ongoing, involving a far larger sample of people, in the U.K., Brazil, and South Africa. A Phase III human trial of the Oxford vaccine involving some 30,000 people will begin in the United States in August.

“While there is more work to be done, today’s data increases our confidence that the vaccine will work and allows us to continue our plans to manufacture the vaccine at scale for broad and equitable access around the world,” Mene Pangalos, Executive Vice President of BioPharmaceuticals Research and Development at AstraZeneca, said.

The U.S. government has invested a whopping $1 billion in Oxford’s trial vaccine already, gambling on it being a success to secure millions of doses as soon as possible. 

Britain has also poured about $90 million into the work by Oxford’s Jenner Institute for vaccine research. On Monday, British government Business Secretary Alok Sharma called the latest trial results “extremely encouraging, taking us one step closer to finding a successful vaccine to protect millions in the U.K. and across the world.”  

The Oxford vaccine has been out in front of about 15 serious global competitors for months, but a few others are hot on the team’s heels.


Moderna advances potential COVID-19 vaccine

Just last week, officials announced similarly hopeful results from Phase I trialing of a vaccine being developed by the National Institutes of Health and Moderna Inc. The experimental vaccine will move to its most important step around the end of July: A 30,000-person study to prove the shots are effective at stopping coronavirus infection.

The U.K. vaccine news was published Monday in The Lancet along with another study on a Chinese vaccine trial. That vaccine also showed promise, creating a strong antibody and T cell response in more than 90 percent of those given the injection after 28 days.

Britain has also poured about $90 million into the work by Oxford’s Jenner Institute for vaccine research. On Monday, British government Business Secretary Alok Sharma called the latest trial results “extremely encouraging, taking us one step closer to finding a successful vaccine to protect millions in the U.K. and across the world.”  

The Oxford vaccine has been out in front of about 15 serious global competitors for months, but a few others are hot on the team’s heels.


Moderna advances potential COVID-19 vaccine

Just last week, officials announced similarly hopeful results from Phase I trialing of a vaccine being developed by the National Institutes of Health and Moderna Inc. The experimental vaccine will move to its most important step around the end of July: A 30,000-person study to prove the shots are effective at stopping coronavirus infection.

The U.K. vaccine news was published Monday in The Lancet along with another study on a Chinese vaccine trial. That vaccine also showed promise, creating a strong antibody and T cell response in more than 90 percent of those given the injection after 28 days.

The coronavirus pandemic is still rapidly spreading across the globe, infecting more than 14.44 million people. With more than 188 countries and territories affected, scientists are racing to develop a vaccine against the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the virus that causes the coronavirus disease (COVID-19).

One of the forerunners in this vaccine race is an adenovirus-vectored vaccine from the University of Oxford, in partnership with AstraZeneca, which started its human trials in May.

In June, the research team reported trial results that revealed a single dose of ChAdOx1 nCoV-19 (AZD1222) induced antigen-specific antibody and T cells responses against the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) in animal models.

The vaccine

The Oxford – AstraZeneca vaccine, the ChAdOx1 nCov-19, is made from a virus called ChAdOx1, a weakened version of a common cold virus or adenovirus that causes infections in chimpanzees.

The vaccine, which is now called the AZD1222 COVID-19, is being developed at the University of Oxford’s Jenner Institute and has been licensed to AstraZeneca.

The virus has been genetically modified so that it cannot replicate in humans.

It was developed by adding genetic material, known as spike glycoprotein, which is expressed on the surface of the coronavirus to the ChAdOx1 virus. The scientists say that the vaccine works by making the body recognize and develop an immune response to the spike glycoprotein, stopping the virus from entering human cells.

In June, the team has started the candidate vaccine’s trial in South Africa and Brazil, with 4,000 and 5,000 participants, respectively.

Encouraging early results

The results of the first human clinical trial of the candidate vaccine will be published later today, July 20, by the journal The Lancet.

“We expect this paper, which is undergoing final editing and preparation, to be published on Monday, July 20, for immediate release,” a spokesperson for the journal said in a statement.

The researchers revealed that the experimental vaccine provides double protection from SARS-CoV-2 after early-stage human trials. The volunteers’ blood samples showed that the vaccine had triggered the body to produce both antibodies and killer T-cells to combat the infection.

Antibodies are proteins that can effectively kill the antigen, in this case, the novel coronavirus. Meanwhile, T cells can kill a virus and the cells it has infected, providing a vital part of the body’s response to viral infections. The results are promising since scientists claim that though the antibodies can fade within months, the T-cells may dwell in the blood longer.

The phase 1 trial that involved about 1,000 British volunteers has been completed, and the results are to be published on July 20 in the journal The Lancet. The team is now preparing for the phase 3 trial, which will recruit tens of thousands of people in the United Kingdom, Brazil, the United States, and South Africa. Alongside the phase 3 trial, the team plans a challenge trial.

Challenge trial

Now, the Oxford vaccine team plans to start tests on volunteers who will be intentionally exposed to the virus in a “challenge trial,” which has been deemed controversial since there is still no approved drug to combat the infection.

In challenge trials, the volunteers recruited are healthy and without any underlying health condition. These volunteers are intentionally exposed to the pathogen in a controlled laboratory setting, which means it can be finished in weeks and requires fewer people than other types of trials.

“We’re hoping to be doing challenge trials by the end of the year. This might be in parallel or might be after the phase three trial is completed. They’re not competing for options, they’re complementary,” Professor Adrian Hill, director of Oxford University’s Jenner Institute, said. He added that the team is now preparing the laboratory on the technical side of the trial. In a couple of months, the team hopes to recruit willing volunteers.

The coronavirus continues to spread across the globe. With many countries starting to reopen their economies and ease lockdown measures, the number of cases is expected to skyrocket in the coming months. The situation highlights the dire need for an effective vaccine to contain the spread of the virus and protect people from getting infected.

The United States remains as the nation with the highest number of infections, reaching 3.76 million with a death toll of more than 140,000. Brazil and India follow with a staggering 2.01 million and 1 million confirmed cases, respectively.

Oxford University’s coronavirus vaccine project leader isn’t as enthusiastic as you want him to be.

Clinical trial results from an experimental coronavirus vaccine AstraZeneca (NYSE:AZN) licensed from Oxford University might not be ready this September as hoped. In an interview with The Telegraph, the director of Oxford’s Jenner Institute, Professor Adrian Hill, said the chances we’ll see meaningful trial results by September are increasingly slim due to successful COVID-19 containment efforts. 

Vaccine math is hard 

In April, Prof. Hill said a million doses could be ready by September if results from ongoing efficacy tests prove ChAdOx1 nCoV-19 is safe and effective. Thanks to successful containment efforts, Hill thinks the number of new infections in the U.K. will be too few to draw any meaningful conclusions at such an early date. Out of around 10,000 people expected to enroll in the trial, the researcher expects less than 50 will catch the virus.

While AstraZeneca can easily solve this problem by expanding the study, we still need to wait and see if new participants injected with the vaccine come down with COVID-19 less often than those randomized to receive a placebo. It’s not unusual to see COVID-19 infections put healthy, athletic people in the hospital or worse. Despite popular suggestions, willingly exposing trial participants to SARS-CoV-2 to speed up the vaccine development process is unethical and illegal in at least a hundred different ways.

Recently, AstraZeneca signed an agreement to provide up to 300 million doses of ChAdOx1 nCoV-19 to the U.S. if it works as intended. The company could receive up to $1.2 billion to assist with the vaccine candidate’s development, including a clinical trial expected to enroll 30,000 participants.

Oxford COVID-19 vaccine candidate protects monkeys in trial

London: An experimental vaccine developed by researchers at Oxford University has shown promise in preventing COVID-19, according to a small trial in six monkeys.

The vaccine candidate, ChAdOx1 nCoV-19, is currently being tested in humans too. Over 1,000 volunteers have participated in the clinical trials.

The results of the trial in six rhesus macaque monkeys were posted on Thursday at the preprint repository bioRxiv.

Rhesus macaque monkeys are considered to be good proxies for how drugs could work in humans as their immune systems are similar, but there is not guarantee that the results would replicate in people.

The researchers found that compared to those monkeys who got exposed to the virus that causes COVID-19 but did not receive the vaccine, the monkeys which received the vaccine had less of the virus in their lungs and airways.

The results showed that the ChAdOx1 nCoV-19 vaccination appears to prevent SARS-CoV-2 pneumonia in rhesus macaques.

Researchers from the US government’s National Institutes of Health (NIH) and from the University of Oxford conducted the trial in the US, the BBC reported.

The researchers at Oxford University who have begun testing their potential vaccine against COVID-19 on a small group of human volunteers also hope to expand the tests soon after getting positive results from the initial trial.

The vaccine candidate for COVID-19 was identified by researchers from the Oxford Vaccine Group and Oxford’s Jenner Institute. The potential upcoming vaccine, ChAdOx1 nCoV-19, is based on an adenovirus vaccine vector and the SARS-CoV-2 spike protein.

By vaccinating with ChAdOx1 nCoV-19, the scientists hope to make the body recognise and develop an immune response to the Spike protein that will help stop the SARS-CoV-2 virus from entering human cells and therefore prevent infection.

Over 100 experimental COVID-19 vaccines are currently in different stages of development.

In the global race to find a vaccine, Oxford University just jumped way ahead of the pack. Human testing is already underway, and scientists say they’re hopeful a coronavirus vaccine will be widely available by September.

Technology the lab had already developed in previous work on inoculations for other viruses, including a close relative of COVID-19, gave it a head start.

“Well personally, I have a high degree of confidence about this vaccine, because it’s technology that I’ve used before,” said Sarah Gilbert, a professor of vaccinology at the university.

The vaccine takes the coronavirus’ genetic material and injects it into a common cold virus that has been neutralized so it cannot spread in people. The modified virus will mimic COVID-19, triggering the immune system to fight off the imposter and providing protection against the real thing.

The experimental vaccine has reportedly worked in protecting rhesus macaque monkeys that were exposed to heavy quantities of COVID-19.

In the human trials, 550 participants are given the vaccine, and another 550 receive a placebo.

“It feels like finally, I am able to do something…” said Oxford scientist and trial volunteer Elisa Granato. “This was a way for me to contribute to the cause.”

Wasting no time, the largest drugmaker in the world, based in India, will start producing millions of the Oxford vaccines by next month, even before they’ve been proven to work.
